Nach einem verheerenden Atomkrieg suchen sechs menschliche Fraktionen im Centauri-System Zuflucht. Am Rande der Ausrottung werden die Menschen von einer überlegenen Zivilisation, angeblich den Centauriern, gerettet, und jede Fraktion erhält einen Neuanfang auf einem neuen Planeten. So scheint der Exodus der Menschheit auf den Planeten, die den Stern Proxima Centauri umkreisen, beendet zu sein. Als die Centaurier ihre Abreise ankündigen, beginnt der Kampf um die Macht erneut.

In Exodus: Proxima Centauri kämpfen die sechs Fraktionen um die Dominanz in einem epischen Imperiumsaufbau-Spiel und streben danach, ein neues menschliches Imperium aus den Überresten der centaurischen Zivilisation zu errichten. Jeder Spieler wird seine eigene Raumflotte aus maßgeschneiderten Schiffen aufbauen und neue Planeten erobern, gegen den centaurischen Widerstand und die anderen Spieler kämpfen, politische Entscheidungen aushandeln und abstimmen, centaurische Technologien erforschen und vieles mehr.
